Musculoskeletal disorders (MSKDs) are a major global health concern, significantly affecting mobility, quality of life, and healthcare systems, yet their complex etiology remains insufficiently understood. Utilizing the UK Biobank’s comprehensive dataset, this study aims to investigate how systemic factors contribute to the onset, progression, and severity of MSKDs, while also considering the influence of genetic predisposition and environmental factors such as lifestyle and behavioral patterns. By integrating polygenic risk scores (PRS) with multi-omics data and imaging modalities, we will explore potential biomarkers and predictive models that can enhance early detection, improve disease classification, and support personalized treatment strategies. Through a deeper understanding of these interactions, our research seeks to inform more effective prevention and intervention approaches, ultimately improving patient outcomes and reducing the global burden of MSKDs.
