Research questions: 1) What genetic variants and modifiable lifestyle factors are associated with venous thromboembolism (VTE), deep vein thrombosis (DVT), arterial thrombosis, and altered coagulation/fibrinolysis traits? 2) Do gene-environment interactions modulate thrombosis risk, and can these inform stratified prevention strategies?
Objectives: Identify novel genetic loci linked to thrombosis subtypes and coagulation traits; quantify associations between lifestyle factors (e.g., obesity, smoking, hormone use) and thrombosis risk; explore gene-lifestyle interactions to refine risk stratification models.
Scientific rationale: Thrombotic disorders (VTE/DVT, arterial thrombosis) are leading causes of morbidity/mortality globally. UK Biobank’s large cohort (500k+ participants) with genomic, clinical, and lifestyle data enables robust analysis of multifactorial determinants. Understanding genetic and environmental drivers will improve early risk identification and inform targeted public health interventions, addressing an unmet clinical need for personalized thrombosis prevention.
