This research aims to explore the relationship between Frailty Index (FI) and cardiovascular diseases (CVD) through a multifaceted approach:
Research on the Association Between FI and CVD:
i) Prospective Cohort Studies: We will conduct studies to investigate the association between FI and various cardiovascular diseases, as well as the comorbidity of FI with CVDs. These studies will track participants over time to assess how FI impacts the onset and progression of CVD.
ii) Single-Sample Mendelian Randomization (MR) Analysis: We will use genetic risk scores to perform MR analysis, examining whether genetic predispositions to FI influence the risk of developing CVD.
Population Stratification Analysis: This analysis will explore how the relationship between FI and CVD varies across different population subgroups, accounting for factors such as age, sex, and ethnicity.
iii) Two-Sample Mendelian Randomization (MR) Analysis: We will apply MR analysis using separate datasets to confirm causal relationships and validate our findings.
Mechanistic Studies on the Comorbidity of FI and CVD:
i) Identification of SNP-Level Loci: We will identify specific single nucleotide polymorphisms (SNPs) associated with FI and CVD to pinpoint genetic variants that may contribute to the comorbidity.
ii) Identification of Risk Genes at the Gene Level: Utilizing MAGMA and eQTL analysis, we will identify risk genes and examine their role in the relationship between FI and CVD.
iii) Functional Enrichment Analysis: This analysis will assess the biological pathways and functional processes involved in the comorbidity of FI and CVD, providing insights into the underlying mechanisms.
